✅ Description:
Doxophylline is a newer bronchodilator belonging to the xanthine group. Structurally, it differs from theophylline due to the presence of a dioxolane group at position 7.
It works by selectively inhibiting phosphodiesterase-4 (PDE4), leading to relaxation of bronchial smooth muscles.
Compared to theophylline, it has lower affinity for adenosine receptors (A1 and A2), which contributes to a better safety profile and fewer side effects.

✔️ Doxophylline is generally well tolerated, but may cause mild symptoms such as:
- Nausea and vomiting
- Headache
- Gastric discomfort
- Heartburn
✔️ Dosage & Administration | Doxophylline
Adults
- 400 mg tablet 2–3 times daily, or as directed by a physician
Elderly
- 200 mg tablet 2–3 times daily
Children
- >12 years: 200 mg tablet or 10 ml syrup 2–3 times daily
- 6–12 years: 6–9 mg/kg body weight twice daily
Sustained Release (SR) Form
- If the total daily dose is 400 mg, SR tablet may be taken once daily
Always follow the physician’s recommendation for dosing.

- Avoid concurrent use with other xanthine derivatives
- May interact with:
- Ephedrine (risk of additive toxicity)
- Troleandomycin, lincomycin, clindamycin
- Allopurinol
- Cimetidine, ranitidine
- Propranolol
- Influenza vaccine
These may reduce liver clearance, increasing drug levels in the blood.
